The German club are keen to add to their strike-force after seeing Mario Gomez leave for Bayern Munich in the summer and thought they had a deal lined up midweek for the Dutch striker.
But Huntelaar failed to respond before the club's deadline of Wednesday evening and so Stuttgart announced that the offer had been withdrawn.
But Madrid's phone has been ringing once again as the Bundesliga side have now asked about Negredo, who is expected to command a similar asking price, Marca state.
Stuttgart claimed that they had made an "extraordinary" offer for Huntelaar and while their new target will also cost €18 million, his wages will not be as high.
Both Huntelaar and Negredo have been linked with moves to the Premier League also and if Stuttgart make their bid for Negredo official then they could see the same thing happen again.
While they were waiting for a response from Huntelaar, it was reported that he and his agent were hoping to hear from Tottenham Hotspur or another interested English club.
Negredo has been linked with Tottenham as well, but Liverpool are also in the market for a powerful striker and could be tempted to ask for him to be included in a swap deal for Xabi Alonso.
